NASA officials were received at Kennedy Space Center—the reason is simple: the US military doesn't have a very high regard for S.H.I.E.L.D.
"Regarding the Avengers' previous proposal to postpone the launch, we recently reached a consensus that we should send an unmanned spacecraft to the target for data collection. If all the data is normal, we will conduct a manned space mission next year."
It seems that the story of a super-powered, psychopathic serial killer spurred NASA to reach a consensus: they should not launch any living people into space this year to avoid any negative consequences.
After all, Spider-Man's information came from a prophecy, and they could easily pretend to be scientists and deny it. But S.H.I.E.L.D.'s information is reality. You can't wait until something goes wrong and then be held accountable for insisting on a manned launch when a superpowered psychopath appears at Cape Canaveral, can you?
Do you expect them to explicitly say that astronauts are not human?
In reality, it might be possible, but this is a novel, and novels are supposed to be logical.
“We’re not here to disrupt your space mission.” Although Coulson was satisfied with the outcome, he still explained his purpose: “Our goal is to apprehend the suspect. Since no serious incidents have occurred at Cape Canaveral recently, it means we’ve arrived in time.
